Recombinant Nucleases

MoBiTec expands its R&D enzyme palette by a number of recombinant nucleases for applications in R&D and in diagnostics.

  • HS-Nuclease, recombinant Endonuclease : HS-Nuclease can be used to reduce viscosity of cell lysates and remove nucleic acid contaminations from sample preparations.

  • Recombinant RNase T1: RNase T1 from Aspergillus oryzae useful for RNA structure and mapping studies, for RNA protection assays, as well as for removal of RNA from DNA samples.

  • Recombinant RNase T2: First recombinant Ribonuclease T2. Available in extremely high purity.

  • Recombinant Exonuclease III: Enzyme useful for nested unidirectional deletions, generation of a single-stranded templates, site-directed mutagenesis and other cloning tasks.

Ribonuclease T1 (RNase T1) is an endoribonuclease that specifically cuts RNA at the 3'-end of guanosine residues through a 2', 3'-cyclic phosphate intermediate mechanism. RNase T1 is cloned and produced recombinantly to create a highly pure enzyme without contaminating DNase or nonspecific RNase activity.
RNase T1 is used for RNA structure and mapping studies and for RNA protection assays, as well as for removal of RNA from DNA samples.


Exonuclease III (ExoIII) of E. coli is a 31 kD monomeric, globular protein combining multiple catalytic activities in one active site. It acts on doublestranded (ds) DNA as a 3'-5' exonuclease, a 3'- phosphomonoesterase, an apurinic/apyrimidimic (AP) sites specific endonuclease and an exonucleolytic ribonuclease H. Of particular interest for molecular biological methods is its exonucleolytic activity, removing 5'-mononucleotides from the 3'-hydroxyl ends of ds DNA, leaving protruding 5'-termini. Its catalytic rate can be adjusted by temperature and NaCl concentration and thus allows for the generation of single-stranded DNA templates for sequencing or recombination methods.
