TubulinTrackerTM Green Reagent
Ideal for highly selective live-cell staining, e.g. of cell structures in live GFP-transfected cells
TubulinTracker Green reagent (T34075), an acetylated version of Oregon Green 488 paclitaxel (Flutax-2, P22310), makes live-cell tubulin staining easier. With greater solubility and better cell permeability than the original paclitaxel, TubulinTracker Green reagent is highly selective for polymerized tubulin and provides more uniform labeling. Initially uncharged and nonfluorescent, TubulinTracker Green reagent easily diffuses through the plasma membrane of live cells. Once inside the cell, the lipophilic blocking group is cleaved by nonspecific esterases, resulting in a green-fluorescent charged paclitaxel.

ER-TRackerTM Green & Red Dyes
For live cell endoplasmatic reticulum staining
The green-fluorescent ER-Tracker Green and red-fluorescent ER-Tracker Red dyes (E34251, E34250) correspond to the drug conjugates BODIPYFL glibenclamide and BODIPY TR glibenclamide, which exhibit excitation/emission maxima of ~540/511 nm and 587/615 nm, respectively. Glibenclamide (glyburide), a drug taken daily by millions of diabetic patients to correct hyperglycemia, has been used to investigate pancreatic-cell activity and insulin secretion and to study myocardial cell function and heart arrhythmia. Glibenclamide binds to the sulphonylurea receptors of ATP-sensitive K+ channels, which are prominent on ER; the pharmacological activity of glibenclamide could potentially affect ER function. Variable expression of sulphonylurea receptors in some specialized cell types may result in non-ER labeling.

SelectFXTM Alexa Fluor® 488 ER Labeling Kit
Allows highly specific, bright labeling of the ER
The SelectFX Alexa Fluor® 488 Endoplasmic Reticulum Labeling Kit (S34200) provides all the reagents required to fix and permeabilize mammalian cells and then specifically label the ER. To achieve ER labeling, this kit employs a primary antibody directed against an ER-associated protein, protein disulfide isomerase (PDI), and an Alexa Fluor® 488 dye-labeled secondary antibody. The Alexa Fluor® 488 dye exhibits bright green fluorescence that is compatible with filters and instrument settings appropriate for fluorescein.
Each kit contains:
- Mouse IgG2b anti-protein disulfide isomerase (PDI) antibody
- Highly cross-adsorbed Alexa Fluor 488 goat anti-mouse IgG antibody
- Concentrated fixative solution

Fixed, permeabilized bovine pulmonary artery endothelial cells visualized using components of the SelectFX™ Nuclear Labeling Kit and Alexa Fluor® phalloidin conjugates. Nuclei and F-actin were stained, respectively, with (top left) DAPI & Alexa Fluor® 680 phalloidin, (top right) SYTOX® Green dye & Alexa Fluor® 568 phalloidin, (bottom left) 7-AAD & Alexa Fluor® 488 phalloidin, and (bottom right) TO-PRO-3 dye & Alexa Fluor® 350 phalloidin. |
Highly sensitive nuclei staining
The SelectFXTM Nuclear Labeling Kit (S33025) provides four spectrally distinct fluorescent dyes - blue-fluorescent DAPI, green-fluorescent SYTOX Green stain, red-fluorescent 7-aminoactinomycin D (7-AAD) and far red-fluorescent TO-PRO-3 dye - for staining nuclei in fixed and permeabilized cells and tissues with essentially no cytoplasmic background staining. When used according to the protocol provided, the dyes in the SelectFXTM Nuclear Labeling Kit provide highly selective nuclear staining with little or no cytoplasmic labeling; they are ideal for use as counterstains in multicolor applications. The stained nuclei stand out in vivid contrast to other fluorescently labeled cell structures when observed by fluorescence microscopy. These dyes have excitation wavelengths that match the common laser lines for confocal microscopy and flow cytometry and can be used with standard filter sets on fluorescence microscopes and microplate readers. The staining protocol provided is compatible with a wide range of cytological labeling techniques, including direct or indirect antibody-based detection methods, mRNA in situ hybridization and staining methods that incorporate organelle- and cytoskeleton-selective fluorescent probes (including MitoTracker mitochondrion-selective probes and Alexa Fluor® dye conjugated phalloidins). The dyes can also be used to fluorescently stain cells for analysis in multicolor flow cytometry experiments. All dyes are provided as stock solutions, convenient for diluting and staining, and each dye is also available separately.
The SelectFX Nuclear Labeling Kit contains:
- DAPI, a blue-fluorescent counterstain (excitation/emission maxima ~358/451 nm)
- SYTOX Green, a green-fluorescent counterstain (excitation/emission maxima ~504/523 nm)
- 7-Aminoactinomycin D (7-AAD), a red-fluorescent counterstain (excitation/emission maxima ~546/647 nm)
- TO-PRO-3 iodide, a far red-fluorescent counterstain (excitation/emission maxima ~642/661 nm)

The SelectFXTM Nuclear Labeling Kit contains sufficient reagents to prepare ~100 assays with each stain at 300 µL per assay.
